Abstract   Measurement of DNA content by flow cytometer was performed in the synchronous colorectal adenoma(s) and adenocarcinoma. This study was performed as part of our projects which were designed to examine the changes of adenomacarcinoma sequence and transitional mucosae near tumor in colorectal cancer. The ploidy and proliferative indices(S-phase fraction, S+ G2M) of 9 synchronous colorectal adenoma(s) and adenocarcinoma were analysed in the tissue samples of serially sectioned transitional mucosae and tumors. 2 cases of adenocarcinomas and 5 cases of adenomas were diploid, while average DNA indices of them were 1.34+-0.41, and 1.19+-0.30 respectively. 15 tissue samples of 1 cm prior tumor and 33 of between tumors were diploid except 2 samples of diploid-cycling in the same case. The average proliferative indices of S-phase and S+G2M were 13.15+-1.50% and 19.08+-7.75% in the adenocarcinomas, while 7.63+-3.97% and ll.ll+4.73% in the adenomas. In the transitional mucosae, proliferative indices near adenocarcinoma between tumors showed the highest value. The proliferative indices of transitional mucosae between single adenoma and adenomas did not show any significant difference. Ciinical stage, location, and differentiation in adenocarcinomas and histologic type, and degree of dysplasia in adenomas did not related to ploidy and proliferative indices of tumors and transitional mucosae. In conclusion, the effect of adenocarcinomas on the transitional mucosae between tumors might be admitted. From the aradual increase of DNA indices in transitional mucosae, adenomas, and adenocarcinomas, it could be interpreted as the possibility of sequential abnormalities of chromosomes in the adenoma-carcinoma sequence in colorectal adenocarcinomas.
